dendritic cells bearing hla-g inhibit t-cell activation in type 1 diabetes

چکیده
hla-g is normally expressed on human trophoblast cells. it is a non-classical mhc molecule class i b. the role of hla-g in diabetic type 1 is not known. we investigated the role of ifn-β in induction hla-g expression on the monocyte derived dendritic cells (dc) in diabetes type 1. treatment of dendritic cell with ifn-β in vitro from diabetic patients (n=20) and normal subjects (n=20) resulted to the production and expression of hla-g on these cells from both groups. however, comparison of dc from the diabetic patients with dc from the controls revealed lower levels of hla-g molecules in dc from diabetic patients. using mixed lymphocyte reaction (mlr), it was found that dc expressing hla-g mediated the inhibition of autologous t cell activation. it is concluded that ifn-β can increase hla-g in dc from diabetic patients; subsequently it may prevent the immune regularly pathway in the diabetic pathogenesis.

متن کاملt-helper cell type-1 transcription factor t-bet is down-regulated in type 1 diabetes
t cells have been identified as key players in the pathogenesis of type 1 diabetes. however, the exact role of t-cell subpopulations in this pathway is presently unknown. the purpose of this study was to assess the expression pattern of two lineage-specifying transcription factors gata - 3 and t - bet , which are important in t helper type 1 (th1) and th2 cell development , respectively. gene e...

متن کاملβig-h3 Represses T-Cell Activation in Type 1 Diabetes.
βig-h3/TGF-βi is a secreted protein capable of binding to both extracellular matrix and cells. Human genetic studies recently revealed that in the tgfbi gene encoding for βig-h3, three single nucleotide polymorphisms were significantly associated with type 1 diabetes (T1D) risk. Pancreatic islets express βig-h3 in physiological conditions, but this expression is reduced in β-cell insult in T1D....
متن کاملAltered Suppressor Function of Regulatory T Cells in Type 1 Diabetes
Background: Type 1 diabetes (T1D) is a T cell mediated autoimmune disease targeting the insulin-producing β cells within pancreatic islets. Autoimmune diseases may develop as a consequence of altered balance between regulatory (Tregs) and autoreactive T cells. Objectives: To evaluate Treg cells frequency and suppressive function in the peripheral blood of newly diagnosed T1D patients in compari...
متن کاملdendritic cell stimulation by ifn-β alters t cell function via modulation of cytokine secretion in diabetes type 1
during antigen capture and processing, mature dendritic cells (dc) express large amounts of peptide-mhc complexes and accessory molecules on their surface. we investigated the role of ifn-β in induction hla-g expression on the monocyte derived dc and cytokine profile in diabetes type 1. we accomplished secretary pattern and total cytokine production of the th1 cytokine (il-2, γifn) and th2 cyto...
